Study the dynamic of shoot on different concentration of 3 types of auxin, most active auxin is IBA. But for in vitro rooting, I must be very cautious to develop a new concentration for rooting medium. Each clone may have a specific requirement for hormon, too high, as I see in E hybrid clones, may develop callus prior to root, which maeans callus differentiate from callus, which I do not preferred. Some literature suspect it as not having vascular connection to stem.
some may even develop root from unwanted parts such as leaves, petiole, which I think not unuseful and can break up easily upon washing.
so we need to test each of them, one by one, within a not too wide/large concentration
