Salinity stress may become a problem in the future, how do we screen tolerant species/clones using in vitro technique. I have some clones, and I want to assesed their salt tolerance.

Michael Dow • If by in vitro technique, you mean growing plants on solid medium on petri dishes, then you should be careful when interpreting your data. Plants do not transpire well when grown on plates and will not accumulate Na+ or Cl- to the same extent compared to growth under transpiring conditions. Therefore the NaCl stress you apply will really be an osmotic stress rather than an ionic stress.

András Ferenc Bálint • The best way is to test "On site", I mean on the field where salinity is a real problem. The results of enzyme activities, tolerance indexes based on hydroponics, etc. showing only a slight correlation with the real tolerance - If you have luck. Otherwise, you will get nice information about your lines (tolerant, sensitive etc.), without any sense.
If your lines have closely the same genetic background (they are different only in one or several key genes), then hydroponics/in vitro techniques are suitable to find the effect of a special gene/mutation. However, if you want to select tolerant lines, you have to use the same conditions as in the field. If you want to check why the tolerant is tolerant/sensitive is sensitive then use the well controlled in vitro techniques to find out the mechanism behind the tolerance.

Tarif Charbaji • The best way to screen plant tolerance to salinity is the overload several doses of NaCl to the media, then observe the reaction of plant to salt stress.
