Sugar In medium Tissue Culture

What if my tissue culture medium minus sugar, will the root still coming out. say I use 1/2MS +IBA 1 + NAA 1 for my eucalyptus micro shoot. Is rooting need external sugar to initiate roots, ?

MARIA FERNANDA PARDO GONZALEZ • hi, in my experience for rooting eucalyptus, i did include sugar at the medium (10gr) but i did not use NAA, with BAP it was enough (but the formula could vary depending the variety - clon).

Saad Javed • rooting might occur without sugar as well, but your shoot may die if their is no alternative carbon source.

Najat Faris • It is the only carbon source for the plants, but 10g/l will be enough , more than that will callus may result. Good luck

Dr. Kuldeep Sharma • During micropropagation a carbon source, undoubtedly sucrose (being more cheap) is used. Usually, 1% to 2% is recommended for rooting. Otherwise, if not used roots may emerge out but the overall growth of the plantlet will arrest due to sugar deprivation.

I have tried rooting in liquid media (1/2MS+auxins) using paper bridges, but in most instances callus formed at junction lowers transplantaion efficiency. With micrografting, transplantaion efficiency is almost 98%. "

Anju Gulati-Sakhuja • Hi Fredy, Micrografting is very good technique for rooting of in vitro regenerated shoots (ref: Gulati A, P Schryer and A McHughen (2001) Regeneration and micrografting of lentil shoots. In Vitro Cellular Dev. Biol.- Plant 37: 798-802). I am confident that this technique will work for eucalyptus.
